UCP3 + UCP1 were immunoprecipitated from 1mg of Mouse skeletal muscle whole cell lysate with ab193470 at 1/50 dilution. Western blot was performed from the immunoprecipitate using ab193470 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/1000 dilution.

Uncoupling proteins UCP3 and UCP1 are members of the mitochondrial anion carrier protein family. These proteins play significant roles in mitochondrial proton leak impacting the production of ATP and regulation of energy balance. UCP1 also known as thermogenin has a molecular mass of approximately 32 kDa and is primarily expressed in brown adipose tissue. In contrast UCP3 is found mostly in skeletal muscle and is slightly larger around 33 kDa. Both proteins facilitate the uncoupling process in mitochondria dissipating proton gradient energy as heat instead of ATP synthesis.
Biological function summary

UCP3 and UCP1 are involved in thermoregulation and energy metabolism. These proteins do not operate as part of a larger protein complex but contribute to distinct cellular processes. UCP1 generates heat in brown adipose tissue enabling thermogenesis a process important for maintaining body temperature in cold environments. UCP3 plays a role in regulating fatty acid metabolism and mitigating the effects of reactive oxygen species within muscle cells. This protein may protect mitochondria from damage due to oxidative stress.

Pathways

The roles UCP3 and UCP1 play intersect significantly with the oxidative phosphorylation and lipid metabolism pathways. UCP1 links closely to the thermogenesis pathway where it aids in converting stored energy in brown fat into heat alongside signaling proteins like PGC-1α. UCP3 integrates into pathways that involve the oxidation of fatty acids working with proteins such as peroxisome proliferator-activated receptors (PPARs) to regulate energy combustion in muscle tissues.

UCP3 and UCP1 have connections to obesity and type 2 diabetes. Low UCP1 activity can result in reduced thermogenesis and increased susceptibility to obesity with leptin and insulin involved in this dysfunctional process. UCP3 variations have associations with impaired fatty acid oxidation contributing to lipid accumulation in muscles and insulin resistance a prominent issue in type 2 diabetes. Understanding the modulation of these proteins offers avenues for therapeutic intervention in metabolic disorders.

Functions as a long-chain fatty acid/LCFA and proton symporter, simultaneously transporting one LCFA and one proton through the inner mitochondrial membrane (PubMed : 24196960, PubMed : 28781081).
