UGT1A4 also known as UDP-glucuronosyltransferase 1A4 is an enzyme that plays a significant role in drug metabolism. It weighs approximately 61 kDa. UGT1A4 catalyzes the glucuronidation process where it transfers glucuronic acid from UDP-glucuronic acid to substrates making them more water-soluble and easier to excrete. This process mainly occurs in the liver where UGT1A4 expression is high but expression is also found in other tissues like the kidney.
Biological function summary

UGT1A4 functions in the detoxification and metabolism of various endogenous and exogenous compounds. It is not a part of a complex. It primarily metabolizes drugs such as antipsychotics and anticonvulsants moving them towards elimination from the body. Isoform 1. UDP-glucuronosyltransferase (UGT) that catalyzes phase II biotransformation reactions in which lipophilic substrates are conjugated with glucuronic acid to increase the metabolite's water solubility, thereby facilitating excretion into either the urine or bile (PubMed : 18177842, PubMed : 24641623). Essential for the elimination and detoxification of drugs, xenobiotics and endogenous compounds (PubMed : 18177842). Involved in the glucuronidation of calcidiol, which is the major circulating form of vitamin D3 essential for the regulation of calcium and phosphate homeostasis (PubMed : 24641623). Also glucuronidates the biologically active form of vitamin D3, calcitriol, probably leading to its biliary transport and intestinal reabsorption (PubMed : 18177842).. Isoform 2. Lacks UDP-glucuronosyltransferase (UGT) activity but acts as a negative regulator of isoform 1.
