UGT3A1 also known as UDP-glucuronosyltransferase 3A1 is an enzyme involved in the process of glucuronidation which adds glucuronic acid to substances aiding in their solubility and excretion from the body. This enzyme has a molecular mass of approximately 60 kDa. UGT3A1 is primarily expressed in the liver with some expression in other tissues such as the kidney and intestine. It acts by conjugating a variety of lipophilic compounds including drugs toxins and endogenous compounds like hormones and bilirubin facilitating their elimination.
Biological function summary

This enzyme plays an important role in detoxification and metabolism processes. UGT3A1 is part of the larger UDP-glucuronosyltransferase family which participates in phase II of drug metabolism. It does not function individually but as a homodimer offering structural support for catalytic activity. By making hydrophobic molecules more water-soluble UGT3A1 reduces their biological activity and assists in their removal from the body maintaining homeostasis.

Pathways

UGT3A1 is integral to the glucuronidation pathway a critical phase II metabolic process. This pathway is essential for the clearance of a wide range of substrates including therapeutic drugs and endogenous substrates. UGT3A1 interacts with the cytochrome P450 family which primarily deal with phase I of drug metabolism. Together they constitute a comprehensive metabolic framework essential for biotransformation of many xenobiotics and endogenous substances.

UGT3A1 has been associated with altered drug metabolism potentially influencing drug clearance and leading to adverse drug reactions. Additionally variations in UGT3A1 activity might contribute to disorders involving hormone imbalances due to its role in hormone glucuronidation. Polymorphisms in UGT3A1 gene may impact its expression and function linking it indirectly with CYP3A4 another pivotal enzyme in drug metabolism affecting overall pharmaceutical efficacy and safety.
