ULK2 also known as UNC-51-like kinase 2 is a serine/threonine-protein kinase positioned within cellular processes. With an approximate molecular mass of 124 kDa ULK2 is expressed in various tissues prominently within the brain heart and skeletal muscle. This protein exhibits kinase activity essential for transferring phosphate groups to specific substrates affecting their function and activity.
Biological function summary

ULK2 functions as an important regulator in cellular autophagy the process that removes unnecessary or damaged cellular components. It acts as part of the ULK complex which includes ATG13 FIP200 and ATG101. This complex initiates the autophagy process by responding to nutrient availability and energy status within the cell effectively managing cellular homeostasis and survival.

Pathways

ULK2 is mainly involved in the autophagy pathway and the mTOR signaling pathway. The mTOR pathway influences cell growth and metabolism in response to nutritional signals hence ULK2 interacts with mTORC1 to relay signals important for appropriate autophagy initiation. Additionally ULK2 shares a functional relationship with AMPK which senses cellular energy levels and modulates autophagy in part through ULK2 activation.

ULK2 is associated with neurodegenerative diseases like Alzheimer's disease and can play a role in the pathology of certain cancers. Aberrant ULK2 activity alters autophagic processes contributing to the accumulation of misfolded proteins and impaired cell survival. The protein interacts with p62/SQSTM1 a known player in neurodegenerative disease pathways underlining its relevance to disease mechanisms.

Target data

Serine/threonine-protein kinase involved in autophagy in response to starvation. Acts upstream of phosphatidylinositol 3-kinase PIK3C3 to regulate the formation of autophagophores, the precursors of autophagosomes. Part of regulatory feedback loops in autophagy : acts both as a downstream effector and a negative regulator of mammalian target of rapamycin complex 1 (mTORC1) via interaction with RPTOR. Activated via phosphorylation by AMPK, also acts as a negative regulator of AMPK through phosphorylation of the AMPK subunits PRKAA1, PRKAB2 and PRKAG1. May phosphorylate ATG13/KIAA0652, FRS2, FRS3 and RPTOR; however such data need additional evidences. Not involved in ammonia-induced autophagy or in autophagic response of cerebellar granule neurons (CGN) to low potassium concentration. Plays a role early in neuronal differentiation and is required for granule cell axon formation : may govern axon formation via Ras-like GTPase signaling and through regulation of the Rab5-mediated endocytic pathways within developing axons.
