Function

Plays a role in vesicle maturation during exocytosis as a target of the diacylglycerol second messenger pathway. Is involved in neurotransmitter release by acting in synaptic vesicle priming prior to vesicle fusion and participates in the activity-depending refilling of readily releasable vesicle pool (RRP) (By similarity). Essential for synaptic vesicle maturation in a subset of excitatory/glutamatergic but not inhibitory/GABA-mediated synapses (By similarity). In collaboration with UNC13A, facilitates neuronal dense core vesicles fusion as well as controls the location and efficiency of their synaptic release (By similarity).

Activity summary

UNC13B also known as Munc13-2 is a large protein with a molecular mass of approximately 220 kDa. It plays a critical role in synaptic vesicle fusion and neurotransmitter release at the presynaptic terminal. UNC13B is expressed in the central nervous system particularly in the brain. The protein acts as a scaffold enabling the proper assembly and function of synaptic vesicle-associated molecular machinery.

Biological function summary

The protein interacts closely with SNARE proteins to prime synaptic vesicles for exocytosis. UNC13B is an important component of the synaptic vesicle release machinery functioning as part of larger molecular complexes involved in synaptic transmission. It facilitates the transition of synaptic vesicles from a fusion-competent state to an active state ready for neurotransmitter release. This interaction positions it as significant for efficient neural communication and plasticity.

Pathways

Synaptic vesicle cycle and neurotransmitter release cascade involve UNC13B. It works together with syntaxin-1 and Munc18-1 as part of the SNARE complex machinery. This interaction ensures the transmission of nerve signals by facilitating neurotransmitter release at synaptic junctions. Proper functioning of this pathway is essential for maintaining normal neural circuit activity.

Associated diseases and disorders

Defects in UNC13B have links to neuropsychiatric conditions like schizophrenia and epilepsy. These diseases often associate with abnormal synaptic transmission and neural communication failures. Research shows that imbalances in the protein's regulatory pathways involving proteins like Synaptotagmin might contribute to these disorders. Understanding UNC13B's precise role might offer insights into potential therapeutic targets for these neural communication-related diseases.
