Flow Cytometric analysis of mouse PBMC (Mouse primary peripheral blood mononuclear cell) treated with 100ng/ml LPS for 16h, labeling uPA Receptor/U-PAR using ab255815 at a 1/600 dilution. Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) was used as the secondary at a 1/2000 dilution.

Cells were stained with rabbit IgG (Left) or ab255815 (Right). Then stained with anti-CD11b conjugated to Pacific blue.

Gated on viable cells.

Expression of uPAR on monocytes and granulocytes are observed in LPS-treated PBMC (PMID : 11447203).

uPA Receptor/U-PAR was immunoprecipitated from 0.35 mg LLC1 (mouse lung carcinoma) whole cell lysate 10μg with ab255815 at 1/30 dilution (2μg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab255815 at a 1/1000 dilution (0.59 μg/ml). VeriBlot for IP Detection Reagent (HRP) (ab131366) was used at a 1/5000 dilution.

Lane 1 : LLC1 (mouse lung carcinoma) whole cell lysate 10μg.

Lane 2 : ab255815 IP in LLC1 whole cell lysate.

Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab255815 in LLC1 whole cell lysate.

Blocking/Dilution buffer : 5% NFDM/TBST.

Exposure time : 3 seconds.

uPAR is a glycoprotein. Full-length GPI-anchored uPAR (60kDa) and truncated soluble uPAR (45kDa) are observed. The molecular weight observed is consistent with what has been described in the literature (PMID : 19667118, PMID 24999729, PMID : 11535128).
