Activity summary

URB2 also known as Utp23 is a protein with an approximate mass of 42 kDa. It functions as a nuclease that is essential for the maturation of small nucleolar ribonucleoprotein (snoRNP) complexes. URB2 is found primarily in the nucleolus where it plays a role in processing and modifying ribosomal RNA (rRNA). This protein is integral for the correct assembly of ribosome subunits impacting the overall efficiency and accuracy of protein synthesis.

Biological function summary

URB2 is involved in the ribosome biogenesis process which is a fundamental cellular activity. It participates as a part of the small-subunit (SSU) processome a large ribonucleoprotein complex responsible for the early steps of processing pre-18S rRNA. Through interaction with other nucleases and RNA helicases within the SSU processome URB2 ensures the precise cleavage of rRNA facilitating the formation of functional ribosomes. This activity is directly linked to cellular growth and proliferation.

Pathways

URB2 plays a significant role in the ribosome biosynthesis pathway. It interacts closely with other proteins such as Utp21 and Utp4 both important for the assemble of the SSU processome. The efficient ribosome synthesis pathway is stringent on the function of URB2 because defects in ribosomal assembly can slow down protein production affecting cellular growth. Additionally URB2 is associated with the nucleolar surveillance pathway which monitors ribosome assembly and manages stress responses in the nucleolus.

Associated diseases and disorders

Altered URB2 function can contribute to ribosomopathies which are disorders resulting from defects in ribosome synthesis. Such defects in URB2 have been linked to conditions like Diamond-Blackfan anemia where dysfunctional ribosomal assembly leads to inadequate red blood cell production. URB2 is also associated with Treacher Collins syndrome through interactions with the TCOF1 protein impacting craniofacial development. Understanding URB2’s role in these conditions might offer insights into novel therapeutic approaches.
