Supplementary information

Urocortin 3 also known as Stresscopin is a peptide with a mass of approximately 5 kDa belonging to the corticotropin-releasing factor (CRF) family. It operates mechanically by binding to specific receptors primarily CRF2 receptors to exert physiological effects. Researchers have observed expression of Urocortin 3 primarily in the hypothalamus and certain peripheral tissues like the adrenal glands and the gastrointestinal tract. This peptide plays a significant role in influencing stress-related responses through these receptor interactions.
Biological function summary

This peptide is involved in modulating stress behavior and homeostasis. By activating CRF2 receptors it plays a role in reducing anxiety and stress contrasting with other CRF family proteins that typically increase stress responses. Urocortin 3 does not exist as a simple isolated protein but interacts as part of a complex signaling network affecting neuroendocrine functions. Its distribution and receptor preference help it to mediate effects distinct from other CRF family members providing a counterbalance in biological stress responses.

Pathways

Urocortin 3 affects the hypothalamic-pituitary-adrenal (HPA) axis and the cardiovascular system. In the HPA axis it regulates the secretion of adrenocorticotropic hormone (ACTH) and modulates cortisol release impacting overall stress hormone levels. Relative proteins in these pathways include CRF1 and CRF2 which enliven different aspects of stress and anxiety regulation. In the cardiovascular system Urocortin 3 has been linked to vasodilation and blood pressure regulation.

Urocortin 3's modulation of stress responses ties it to conditions like anxiety disorders and heart disease. In anxiety disorders its role as an anxiolytic contrasts with CRF1 receptor pathways often linked to heightened stress. It's important in maintaining balanced stress responses through CRF2 pathways. In heart disease researchers study its vasodilatory properties for potential therapeutic benefits associating its effects with the regulation of blood pressure and cardiac stress protection in relation to its interaction with CRF2 receptors.
