IHC image of URP2/Kindlin-3 staining in Human normal spleen formalin fixed paraffin embedded tissue section, performed on a Leica Bond™ system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab173416, 0.1μg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

URP2 also known as Kindlin-3 is a protein involved in integrin signaling and cell adhesion processes. It plays a critical role in facilitating the linkage between integrins and the actin cytoskeleton enhancing the signaling cascades that promote cellular adhesion and motility. Kindlin-3 has a molecular mass of approximately 75 kDa. This protein is ubiquitously expressed but found in particularly high levels in hematopoietic cells including leukocytes and platelets.
Biological function summary

Kindlin-3 is essential for the activation and stabilization of integrin complexes which are important for the processes of cell migration adhesion and immune response. It works as a part of the integrin adhesion complex interacting with other cytoplasmic proteins to ensure proper integrin function. Disturbance in Kindlin-3's activity can lead to impaired cell adhesion and migration underlining its importance in both normal cellular functions and pathological conditions.

Pathways

Kindlin-3 plays a significant role in the integrin and focal adhesion signaling pathways. Its interactions within these pathways link it closely with proteins like talin and vinculin which coordinate the signaling transmissions required for cellular movement and environmental interaction. Kindlin-3's ability to regulate integrin activity allows it to participate in cellular processes important for tissue repair and immune responses.

Deficiencies or mutations in Kindlin-3 have strong associations with conditions such as Leukocyte Adhesion Deficiency III (LAD-III) and Glanzmann thrombasthenia-like bleeding disorder. These conditions arise from the impaired integrin activation affecting leukocyte recruitment and platelet aggregation. The protein's role in these disorders highlights its connection with proteins like integrins which fail to function properly in the absence of normal Kindlin-3 activity leading to clinical manifestations in affected individuals.
