US3 also known as UL36 kinase or serine/threonine-protein kinase US3 is a protein encoded by the US3 gene of the herpes simplex virus (HSV). It has a molecular mass of about 45 kDa. This protein plays an important role in kinase activity where it phosphorylates downstream substrates to control cell survival apoptosis inhibition and viral egress. US3 is mainly expressed in cells infected with the herpes simplex virus and it can be found in both the cytoplasm and the nucleus of these cells.
Biological function summary

US3 functions to modulate host cell processes to benefit viral replication. It acts as a kinase and is part of the viral tegument a complex involved in viral assembly and stability. By altering host cell signaling pathways US3 can prevent premature apoptosis providing an environment for efficient viral replication and assembly. This modulation helps maintain cell viability during infection giving the virus sufficient time to replicate.

US3 protein modulates key cellular pathways like the PI3K/Akt and MAPK pathways. The activation of these pathways helps the virus avoid host immune responses. In the PI3K/Akt pathway US3 enhances cell survival by phosphorylating Bad a pro-apoptotic factor. In the MAPK pathway it influences signaling that impacts cell cycle progression. Other related proteins such as Akt and Bad are directly affected by US3's activity linking it to these critical regulatory pathways.

HSV infections involving US3 can contribute to the development of conditions like herpes encephalitis and keratitis. US3's role in blocking apoptosis may increase viral persistence which can worsen these diseases. Proteins like Akt and Bad which interact with US3 during these viral infections are also important because their regulation or dysregulation can influence disease severity and progression.

Multifunctional serine/threonine kinase that plays a role in several processes including egress of virus particles from the nucleus, modulation of the actin cytoskeleton and inhibition of host immune response (PubMed : 25907557, PubMed : 31249678, PubMed : 34935440). Phosphorylates UL31 and UL34, two critical regulators of capsid budding from nucleus to endoplasmic reticulum, thereby facilitating virion egress (PubMed : 15994828, PubMed : 1656069). Modulates and redistributes host components of the nuclear envelope, including LMNA, emerin/EMD and the nuclear matrix protein MATR3 (PubMed : 20962082). In turn, facilitates nuclear pore impairment and capsid release through impaired nuclear envelope (PubMed : 31249678). Phosphorylates envelope glycoprotein B (gB), probably to direct it to the cell surface (PubMed : 19846518). Promotes virus intracellular spread by restructuring host cell cytoskeleton. Blocks host apoptosis to extend cell survival and allow efficient viral replication. Promotes viral gene expression by phosphorylating host HDAC2 to reduce viral genome silencing (PubMed : 20660201). Strongly inhibits TCR-activated signal transduction in T-cells by reducing the ubiquitination of LAT and TRAF6, leading to a suboptimal activation of LAT (PubMed : 25907557). Subverts host antiviral innate immunity by inhibiting type I interferon production through hyperphosphorylation of beta-catenin/CTNNB1 (PubMed : 31801859). In addition, phosphorylates the RNA sensor RIGI and the transcription factor IRF3 to prevent the RLR-mediated antiviral signaling pathway (PubMed : 24049179, PubMed : 34935440). Hyperphosphorylates host RELA and thereby dampens NF-kappa-B signaling (PubMed : 24807716).
