USO1 also known as p115 is a heavy protein weighing approximately 112 kDa. It functions as a general vesicle tethering factor critical for the fusion of Golgi-derived vesicles. USO1 operates by binding to both vesicle and target membranes facilitating vesicular trafficking within the Golgi apparatus. It expresses broadly across various cell types underpinning its essential role in intracellular transport processes.
The USO1 protein plays an instrumental role in maintaining Golgi structure and function. It belongs to the transport protein particle (TRAPP) complex and works closely with several other proteins in the vesicle tethering and fusion steps. By interacting with the Golgi SNAREs USO1 helps mediate the fusion process necessary for protein and lipid transport.
USO1 integrates into cellular trafficking and transport pathways. It participates in the Golgi transport pathway working alongside proteins like GM130 and NSF to ensure stabilized vesicle docking and fusion. USO1's function connects it to the secretory pathway where it assists in uniting the endoplasmic reticulum's production line with the Golgi's packaging and shipping center.
Abnormalities in USO1 function associate with neurological disorders and cancer progressions. USO1's interaction with GM130 links its dysfunctional activity to neuronal migration defects. Additionally aberrations in USO1 levels can influence cancer cell proliferation denoting its potential as a target for therapeutic strategies.
