Activity summary

USP24 also known as Ubiquitin Specific Peptidase 24 is a deubiquitinating enzyme with a molecular mass of approximately 291 kDa. This protein plays a role in the removal of ubiquitin from specific protein substrates which regulates their degradation in the proteasome. It involves the cleavage of the ubiquitin moieties and alters the stability or activity of proteins. USP24 shows expression in several tissues with notable presence in the brain and testis. The specificity of USP24 allows it to finely adjust cellular protein turnover affecting both normal cellular function and stress responses.

Biological function summary

The regulation of protein ubiquitination/deubiquitination is fundamental in maintaining cellular homeostasis. USP24 acts within this regulatory landscape to control pathways involved in cell cycle progression and DNA damage response.     Immunofluorescence staining of HepG2 (Human liver hepatocellular carcinoma cell line) cells with ab314024 at 1:50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor® 569-conjugated Goat Anti-Rabbit IgG(H+L).

    Overlay Peak curve showing HepG2 (Human liver hepatocellular carcinoma cell line) cells stained with ab314024 (pink line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1x10E6 cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG (H+L) at 1:200 dilution for 35min at 4℃. Control antibody (green line) was rabbit IgG (1ug/1x10E6 cells) used under the same conditions. Acquisition of >10,000 events was performed.
