Supplementary information

USP46 also known as Deubiquitinating enzyme 46 is an important player in the process of removing ubiquitin molecules from proteins. This enzyme belonging to the ubiquitin-specific protease family has a molecular mass of approximately 41 kDa. USP46 is widely expressed in various human tissues but significant levels are observed in the brain indicating its importance in neurological processes. Its deubiquitination activity modulate protein turn-over stability and signaling pathways influencing diverse cellular functions.
Biological function summary

USP46 modulates the ubiquitin-proteasome system by removing ubiquitin from substrate proteins which impacts their degradation or activity. It exists as part of a larger protein complex interacting with proteins such as WDR20 which enhance its activity. This interaction allows USP46 to regulate protein homeostasis within neurons and other cells. In the nervous system its activity supports synaptic function and plasticity contributing to effective neural communication and cognitive processes.

Pathways

USP46 mainly acts within the ubiquitin-proteasome pathway an essential mechanism for protein degradation and regulation. It interacts with proteins like UBE3A and UCH-L1 which are important for maintaining protein balance and cellular stress responses. Another important pathway involving USP46 is the neural plasticity and signaling pathway where it helps in regulating the degradation of synaptic proteins contributing to learning and memory.

USP46 has been linked to neurological disorders such as depression and schizophrenia. Abnormalities in its function or expression levels could disrupt normal synaptic activity and protein homeostasis. The enzyme shows a connection with proteins like GSK3β and DISC1 which are implicated in these disorders. Targeting USP46 activity holds potential in developing new therapeutic strategies for such neuro-psychiatric conditions offering avenues for further research and innovation in medical science.

Target data

Deubiquitinating enzyme that plays a role in behavior, possibly by regulating GABA action. May act by mediating the deubiquitination of GAD1/GAD67 (By similarity). Has almost no deubiquitinating activity by itself and requires the interaction with WDR48 to have a high activity (PubMed : 19075014, PubMed : 26388029). Not involved in deubiquitination of monoubiquitinated FANCD2 (PubMed : 19075014).
