Biological function summary
The function of USP47 focuses on maintaining levels of key proteins that require deubiquitination. It stabilizes beta-Catenin and other regulatory proteins by protecting them from proteasomal degradation. As a part of the larger ubiquitin-proteasome system USP47 works with other deubiquitinating enzymes but forms no stable complex with them. Its action ensures the regulation of protein homeostasis impacting cell cycle progression and DNA damage response.
Pathways
USP47 interacts with the Wnt signaling pathway and the DNA damage repair pathway. By controlling the stability of beta-Catenin USP47 influences the Wnt signaling which plays a role in cell growth and differentiation. In the context of DNA repair USP47 interacts with other proteins like Flap endonuclease 1 (FEN1) to contribute to genome integrity by processing DNA lesions and maintaining genomic stability.
