Deubiquitinase with an ATP-independent isopeptidase activity, cleaving at the C-terminus of the ubiquitin moiety. Catalyzes its own deubiquitination. In vitro, isoform 2, but not isoform 3, shows deubiquitinating activity. Promotes plasma membrane localization of ARF6 and selectively regulates ARF6-dependent endocytic protein trafficking. Is able to initiate tumorigenesis by inducing the production of matrix metalloproteinases following NF-kappa-B activation. May act as a GTPase-activating protein for RAB3A (PubMed:19077034).

The USP6 protein also known as Tre-2 or Ubiquitin-Specific Peptidase 6 serves as a deubiquitinating enzyme. It has a molecular mass of approximately 140 kDa. USP6 catalyzes the removal of ubiquitin from specific substrates impacting their stability and degradation. You can find USP6 expression in a variety of tissues but it primarily shows in skeletal muscle and brain tissues.
USP6 regulates numerous cellular processes by modifying protein ubiquitination levels. This protein often interacts with components of the endosomal sorting complex required for transport (ESCRT) playing a part in the regulation of membrane trafficking. Additionally USP6 influences cell signaling pathways by modulating the turnover of receptor proteins important for cell communication.
Several cellular networks involve USP6 that control protein degradation and signal transduction. In particular it plays a role in the Wnt signaling pathway impacting cell proliferation and differentiation. The protein also associates with β-catenin through this pathway affecting cellular processes such as transcriptional activation and cell cycle regulation.
Aberrant USP6 expression links with various conditions. It has a connection to aneurysmal bone cysts where its upregulation results in tumor formation due to enhanced osteoclast activity. The USP6 gene fusion with another protein CDH11 is implicated in this disorder altering normal cellular signaling. Furthermore USP6's dysregulation can also relate to certain soft tissue tumors indicating its role in tumorigenesis.
