Supplementary information

VAMP5 also known as Myobrevin belongs to the vesicle-associated membrane protein family and plays an important role in vesicular transport. This protein is approximately 11 kDa in mass and primarily locates in muscle tissues. VAMP5 is essential in neurotransmitter release and exocytosis processes assisting the fusion of vesicles with target membranes to deliver contained cargo to specific destinations inside the cell. Its expression is higher in skeletal and cardiac muscles reflecting its specialized functions in these areas.
Biological function summary

Vesicle fusion and neurotransmission heavily depend on VAMP5 as it forms part of the SNARE complex. It binds with other SNARE proteins such as SNAP25 and syntaxin to facilitate the docking and fusion of vesicles at the plasma membrane. The correct function of VAMP5 ensures proper communication between cells helping regulate muscle contraction and relaxation. VAMP5's role in muscular tissues emphasizes its importance in maintaining normal physiological processes in these tissues and organs.

Pathways

VAMP5 operates within the exocytosis pathway and is central to synaptic vesicle fusion. It works in concert with other proteins of the SNARE complex to mediate vesicle targeting to the membrane important for neurotransmitter release at synaptic junctions. Additionally it interacts with synaptotagmins which act as sensors for calcium ions further ensuring precise timing and regulation of vesicle fusion during neurotransmission. These interactions illustrate the integration of VAMP5 in critical pathways responsible for neural and muscular functions.

Muscle dysfunction and certain neuropathic conditions relate to VAMP5 abnormalities. Mutations or misregulation in VAMP5 have been implicated in disorders such as myopathy where muscle weakness and degeneration occur. Its connection to the SNARE complex including proteins like SNAP25 highlights the potential for disruptions in this network to contribute to disease pathologies. Understanding VAMP5's role in these disorders can provide insights into therapeutic strategies targeting these conditions.
