VANGL1 also known as Van Gogh-Like Protein 1 plays a mechanical role in cell biology related to polarized cell movement and development. It has a molecular mass of approximately 65 kDa. VANGL1 is expressed in various tissues with significant presence in the kidneys lungs and brain. It localizes to the cell membrane where it participates in the establishment of cell polarity.
VANGL1 is an essential component of the planar cell polarity (PCP) signaling pathway where it guides cell orientation and tissue architecture. This protein is part of a larger protein complex that includes Prickle and Fluorescent in situ hybridization (Fzd) receptors. VANGL1's interactions in this complex facilitate cellular orientation playing a central role in the morphogenesis of epithelial tissues.
Researchers have linked VANGL1 to the Wnt/PCP and non-canonical Wnt signaling pathways. These pathways are important for the regulation of cytoskeletal dynamics and cell motility. VANGL1 interacts with proteins such as the Dishevelled protein family and CELSR1 which are important for signal transduction within these pathways. Proper VANGL1 function ensures accurate cell positioning and movement which are key for embryonic development and organogenesis.
VANGL1 has been implicated in conditions such as neural tube defects and colorectal cancer. In neural tube defects mutations in VANGL1 may disrupt normal cell polarity processes interfering with neural tube closure. Similarly aberrant VANGL1 expression and its interactions with other proteins like KIF7 in the PCP pathway have been associated with the progression of colorectal cancer. Understanding VANGL1's interactions and mutations can provide insights into mechanisms underlying these conditions.
