Supplementary information

VASA also known as DDX4 is a DEAD-box RNA helicase enzyme weighing approximately 79 kDa. VASA is expressed mainly in germ cells including those within the testes and ovaries. Mechanically it unwinds RNA secondary structures facilitating processes like translation and stabilization of RNA molecules. Its activity influences the process of germ cell differentiation by remodeling ribonucleoprotein complexes necessary for proper RNA metabolism.
Biological function summary

The protein plays an essential role in germ cell development and maintenance. VASA is part of the complex involved in ribonucleoprotein processing and it interacts closely with other proteins to regulate RNA dynamics in germ cells. It is fundamental for gametogenesis ensuring the production and maturation of both sperm and eggs. Research shows VASA activity directly affects meiotic chromosome segregation an important feature of reproductive biology.

Pathways

VASA integrates into the P granule pathways interacting with various components critical for germ line specification. It also intersects with the PIWI-interacting RNA (piRNA) pathways which prevent transposable element expression important for safeguarding genome stability. VASA's role alongside proteins like PIWI and AGO proteins illustrates its regulatory role in protecting germ cell genome integrity during development.

VASA's dysfunction relates strongly to infertility and germ cell tumors. Mutations or altered expression levels of VASA have implications in disorders such as spermatogenic failure and ovarian dysgenesis. Research also connects VASA with other proteins like DAZL and NANOS highlighting its involvement in maintaining germ cell homeostasis and avoidance of tumorigenic transformations.
