VEGF or Vascular Endothelial Growth Factor is a signaling protein that plays an essential role in blood vessel formation. Known also as VEGFA its molecular mass is approximately 45 kDa. VEGF is expressed in a variety of tissues especially those with high angiogenic activity such as endothelial cells macrophages and platelets. This protein activates receptors on the surface of endothelial cells triggering a cascade of intracellular events that lead to cellular proliferation migration and increased vascular permeability.
Biological function summary

This protein regulates angiogenesis the process through which new blood vessels form from pre-existing vasculature. It interacts with specific tyrosine kinase receptors such as VEGFR-1 and VEGFR-2. VEGF is not part of a multi-protein complex but it can form homodimers which significantly enhances its activity. It ensures proper vascularization during development and wound healing by directing the growth and branching of vessels. This makes it an important factor in supplying oxygen and nutrients to tissues.

Pathways

VEGF is primarily involved in the VEGF signaling and PI3K/AKT pathways. These pathways allow it to mediate endothelial cell function and survival. In the VEGF signaling pathway it associates closely with members like VEGFR-2 and PLCγ1 which activate further signal transduction needed for angiogenesis. The PI3K/AKT pathway linked with cell proliferation and survival also interacts with VEGF connecting it to other important proteins like AKT and mTOR highlighting its significant role in cellular responses to environmental stimuli.

VEGF has a strong association with cancer and age-related macular degeneration. In cancer overexpression of VEGF leads to pathological angiogenesis providing tumors with the blood supply needed for growth and metastasis. Tumor cells through hypoxia-inducible factors (HIF) upregulate VEGF expression correlating with cancer progression. In age-related macular degeneration abnormal blood vessel growth influenced by VEGF contributes to visual impairment and blindness further implicating the protein in ocular diseases. Targeted therapies often aim at inhibiting VEGF activity to control the disease progression.
