Multifunctional transporter that transports L-glutamate as well as multiple ions such as chloride, sodium and phosphate (PubMed:12151341, PubMed:33440152). At the synaptic vesicle membrane, mainly functions as an uniporter that mediates the uptake of L-glutamate into synaptic vesicles at presynaptic nerve terminals of excitatory neural cells (PubMed:12151341). The L-glutamate uniporter activity is electrogenic and is driven by the proton electrochemical gradient, mainly by the electrical gradient established by the vacuolar H(+)-ATPase across the synaptic vesicle membrane (PubMed:12151341). In addition, functions as a chloride channel that allows a chloride permeation through the synaptic vesicle membrane that affects the proton electrochemical gradient and promotes synaptic vesicles acidification (By similarity). At the plasma membrane, following exocytosis, functions as a symporter of Na(+) and phosphate from the extracellular space to the cytoplasm allowing synaptic phosphate homeostasis regulation (Probable). The symporter activity is electrogenic (PubMed:33440152). Moreover, operates synergistically with SLC18A3/VACHT under a constant H(+) gradient, thereby allowing striatal vesicular acetylcholine uptake (By similarity).

VGLUT3 also known as Vesicular Glutamate Transporter 3 or SLC17A8 functions as a transporter responsible for loading the neurotransmitter glutamate into synaptic vesicles in neurons. It has a molecular mass of approximately 65 kDa. VGLUT3 expression occurs in specific regions of the central nervous system such as the hippocampus and striatum as well as in sensory regions like the cochlea. This transporter lacks the abundance compared to other VGLUTs making its presence more strategically targeted in its roles.
VGLUT3 modulates neurotransmitter signaling by facilitating glutamate release in both excitatory and modulatory circuits. It operates not strictly as part of a homomeric complex but interacts closely with other synaptic proteins to regulate neurotransmission. VGLUT3 presence in serotonin and dopamine neurons indicates its role in modulating their transmission suggesting a diverse function beyond typical excitatory neurotransmitter release. This transporter's activity influences synaptic plasticity and firing patterns vital for maintaining neural circuit balance.
VGLUT3 integrates into the glutamatergic transmission pathway and the modulation of dopaminergic and serotonergic pathways. It works alongside VGLUT1 and VGLUT2 though each has a different regional brain expression allowing diverse neurotransmitter interactions in neural signaling. The transporter interacts with synaptic machinery influencing pathways responsible for synaptic vesicle recycling and neurotransmitter homeostasis ensuring effective transmission functions in responsive neurons.
VGLUT3 influences conditions such as epilepsy and schizophrenia. Immunocytochemistry/Immunofluorescence analysis of SK-N-BE (Human neuroblastoma cell line) cells labeling VGLUT3 using ab134310 at 1/100 for 60 minutes at RT.
