Competitive ELISA - Anti-Vitamin D2+Vitamin D3 antibody [EPPTX-R6-74-3] (ab229821).
Goat anti-rabbit IgG was coated onto 96-wells. Serial dilutions (0, 2.5, 5, 10, 25, 50, 100 ng/mL) of 25-OH-Vitamin-D (25 μL) were incubated with 30-35 ng/mL of ab229821 (50 μL) for 30 minutes by shaking.
This was followed by adding HRP-conjugated 25-OH-Vitamin-D (50 μL) into each well and incubating for another 30 minutes. After washing, 125 μL Tetramethylbenzidine (TMB) substrate was added and incubated for 30 minutes without shaking for color development. OD was read at 450nm within 5 minutes.
