Isoform 1. Electrogenic antiporter that exchanges one cationic monoamine with two intravesicular protons across the membrane of secretory and synaptic vesicles. Uses the electrochemical proton gradient established by the V-type proton-pump ATPase to accumulate high concentrations of monoamines inside the vesicles prior to their release via exocytosis. Transports catecholamines and indolamines with higher affinity for serotonin (PubMed:16326835, PubMed:23337945, PubMed:8643547). Regulates the transvesicular monoaminergic gradient that determines the quantal size. Mediates presynaptic monoaminergic vesicle transport in the amygdala and prefrontal brain regions related with emotion processing in response to environmental stimuli (PubMed:23337945). Isoform 2. Unable to uptake serotonin.

Activity summary

Vesicular monoamine transporter 1 (VMAT1) also known as solute carrier family 18 member 1 (SLC18A1) facilitates the transport of monoamines—such as dopamine serotonin and norepinephrine—into synaptic vesicles. VMAT1 with a molecular weight of approximately 46 kDa is particularly expressed in neuroendocrine cells. These are present in tissues like the adrenal medulla and various parts of the central nervous system. Its transporter function is critical for monoamine storage and release essentially influencing neurotransmitter recycling.

Biological function summary

VMAT1 is involved in the modulation of neural signaling processes. It functions as a monoamine transporter by regulating neurotransmitter levels within vesicles preparing them for secretion. By doing so it influences synaptic transmission and signal transduction impacting mood cognition and other neural activities.
