Supplementary information

WDR16 also known as WD repeat domain 16 is a protein that is involved in several cellular processes through its structural role. It has a molecular mass of approximately 47 kDa and displays expression in various human tissues including the lungs brain and kidney. The protein belongs to the WD repeat protein family characterized by distinct WD repeat structures that facilitate protein-protein interactions.
Biological function summary

WDR16 contributes to important cellular functions and it often participates as part of multiprotein complexes. The protein shows involvement in the regulation of ciliary functions aiding in the proper formation and maintenance of cilia. This role makes it significant in supporting cells' roles in movement fluid flow and sensory functions. Through this biological involvement WDR16 helps ensure proper communication and signaling within cells that contain cilia.

Pathways

WDR16 plays a part in ciliary assembly and maintenance critical for the ciliogenesis pathway. This pathway involves several other proteins such as IFT20 which helps in the transport of ciliary components. Additionally WDR16 has interactions in the Wnt signaling pathway although its specific mechanisms are less thoroughly explored compared to its role in ciliary processes. These pathways emphasize WDR16's role in maintaining cellular structure and signaling required for normal cell function.

Mutations or dysfunction of WDR16 link to ciliopathies a group of disorders caused by defects in cilia structure and function. One such disease is nephronophthisis a kidney disorder characterized by inflammation and fibrosis. In this context WDR16 can connect with proteins like NEK8 which are implicated in similar pathways and disorders. Understanding these connections helps provide insight into the roles of WDR16 in disease pathology and potential therapeutic targets.

Target data

Microtubule inner protein (MIP) part of the dynein-decorated doublet microtubules (DMTs) in cilia axoneme (PubMed : 36191189). Important for proper ciliary and flagellar beating. May act in cooperation with CFAP45 and axonemal dynein subunit DNAH11 (PubMed : 33139725). May play a role in cell growth and/or survival (PubMed : 15967112).
