Supplementary information

WDR72 also known as WD repeat domain 72 is a protein that mechanically functions as a scaffolding unit. This protein enables the assembly of multi-protein complexes essential for maintaining cellular homeostasis. WDR72 has a molecular mass of approximately 110 kDa. It is expressed widely in tissues with significant levels found in the kidneys liver and the brain. This distribution suggests its importance in diverse bodily processes.
Biological function summary

Four roles that the WDR72 protein is involved in include processes related to enamel formation cellular signaling and vesicle transport. WDR72 contributes as a part of a protein complex that regulates ameloblast differentiation which directly impacts enamel development. By facilitating vesicle movement within cells it helps maintain proper cellular activity particularly in the matrix formation of dental tissues.

Pathways

Four key contributions in cellular signaling and enamel development rely on WDR72 functionality. It plays a role in the extracellular signal-regulated kinase (ERK) pathway important for transmitting signals from the cell membrane to the nucleus affecting cell division and survival. WDR72 is linked with proteins such as AMEL and ENAM which are significant within the pathway regulating tooth mineralization and calcification processes essential for strong enamel formation.

Four significant connections exist between WDR72 mutations and amelogenesis imperfecta a hereditary condition marked by defective enamel formation. This connection implicates WDR72 in dental health and integrity. Additionally malfunction or aberrations in WDR72 can relate to renal pathologies due to its expression in kidneys. Within the context of amelogenesis imperfecta WDR72 is connected with AMBN an ameloblast-associated protein highlighting their interaction during enamel formation and pathology when disrupted.

Target data

Plays a major role in formation of tooth enamel (PubMed : 19853237, PubMed : 25008349). Specifically required during the maturation phase of amelogenesis for normal formation of the enamel matrix and clearance of enamel proteins. May be involved in localization of the calcium transporter SLC24A4 to the ameloblast cell membrane.
