Functions as a negative regulator of the PI3 kinase/PI3K activity associated with endosomal membranes via BECN1, a core subunit of the PI3K complex. By modifying the phosphatidylinositol 3-phosphate/PtdInsP3 content of endosomal membranes may regulate endosome fusion, recycling, sorting and early to late endosome transport (PubMed:26783301). It is for instance, required for the delivery of cargos like BST2/tetherin from early to late endosome and thereby participates indirectly to their degradation by the lysosome (PubMed:27126989). May play a role in meiosis (By similarity).

The WDR91 protein also known as WD repeat domain 91 functions mechanistically by interacting with phosphatidylinositol 3-phosphate a lipid important for cellular processes. WDR91 has approximately a molecular weight of 90 kDa. It is expressed in various tissues with high levels found in the brain where it plays a significant role in cellular mechanisms. It contains WD40 repeats known for facilitating protein-protein interactions which are vital for its function in cellular processes.
WDR91 facilitates endosomal signaling and trafficking processes. It assists in the proper sorting and recycling of receptors that regulate cellular responses. WDR91 forms part of a protein complex that includes key players like PIKFYVE which influences lipid synthesis on endosomal membranes. This involvement highlights its role in maintaining balanced cellular responses to environmental signals impacting overall cellular health.
The activities of WDR91 integrate into the endosomal sorting and phosphoinositide signaling pathways. In the endosomal sorting pathway it collaborates with proteins like VPS34 and VPS15 to regulate endosome maturation. Through its function in phosphoinositide signaling it impacts aspects of cellular metabolism and organization affecting pathways linked to cellular growth and survival.
WDR91 is related to a few neurological conditions. Mutations or dysregulation of WDR91 contributes to neurodegenerative disorders such as Alzheimer’s Disease.
