ab174294 staining WDTC1 in HepG2 (Human hepatocellular carcinoma epithelial cell) cells by Immunocytochemistry/Immunofluorescence. Cells were fixed with 4% paraformaldehyde and permeabilized n 0.1% TritonX-100. Samples were incubated with primary antibody at 1/50 dilution (1.6 μg/ml). An AlexaFluor®488 Goat anti-Rabbit (ab150077) was used as a secondary antibody at 1/1000 dilution (2 μg/ml. An AlexaFluor® 594 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker, ab195889 was used as a counterstain antibody (1/200, 2.5 μg/ml). DAPI was used as a nuclear counterstain. WDTC1 also known as Adipose is a protein that plays a role in various cellular processes. It is encoded by the WDTC1 gene and has a molecular weight of approximately 67 kDa. WDTC1 displays expression across many tissues with notable presence in adipose tissue and skeletal muscle. The protein contains WD40 repeats and a TBC (Tre-2/Bub2/Cdc16) domain which assist in protein-protein interactions and cellular trafficking.
Biological function summary

WDTC1 influences energy regulation and fat metabolism. It forms part of a protein complex that includes CUL4-DDB1 contributing to the ubiquitin-proteasome system. This activity regulates degradation of selective proteins ultimately controlling fat storage and mobilization. WDTC1 affects adipogenesis by modulating transcription factors and is involved in lipid metabolism through interactions with other cellular proteins.

Pathways

WDTC1 is actively involved in the regulation of metabolic pathways. It engages with the insulin signaling pathway and the PPARγ (peroxisome proliferator-activated receptor gamma) pathway both essential for energy homeostasis and fat cell differentiation. WDTC1 interacts with insulin receptor substrate proteins and partners with other ligase components to mediate the effects of these pathways on metabolism.

WDTC1 connects to obesity and type 2 diabetes. Variations in WDTC1 expression and function can disrupt normal fat metabolism contributing to these metabolic disorders. It associates with the hormone leptin a critical player in maintaining body weight linking WDTC1 to adipose tissue dysfunction. Understanding the role of WDTC1 may offer insight into therapeutic targets for metabolic disease management.
