WIF1 also known as Wnt inhibitory factor 1 encodes a protein that interacts with Wnt proteins and inhibits their activity. This protein has an approximate molecular mass of 40 kDa and is secreted into the extracellular space. It shows expression in various tissues including the brain lung and kidney. The structure of WIF1 enables it to bind directly to Wnt proteins modulating their signaling capabilities.
Biological function summary

WIF1 participates in the regulation of Wnt signaling which is a critical pathway for cell development and differentiation. It functions to maintain cellular homeostasis by preventing excessive Wnt signaling an essential process in embryonic development and adult tissue maintenance. WIF1 does not form a complex but interacts with Wnt proteins in the extracellular environment ensuring that Wnt signaling remains controlled.

WIF1 plays a significant role in the Wnt signaling pathway which is essential for processes like cell fate determination and proliferation. By inhibiting Wnt activity WIF1 supports maintaining balanced signaling levels within the pathway. It connects to other components like β-catenin and can influence pathways like the Wnt/β-catenin signaling pathway which involves the accumulation and transcriptional activity of β-catenin in the cell nucleus.

WIF1 has been implicated in cancer and osteoarthritis due to its regulatory effect on Wnt signaling. Downregulation of WIF1 often associates with increased Wnt activity contributing to tumor progression in various cancers through interactions with proteins like DKK1 a known Wnt pathway inhibitor. In osteoarthritis altered WIF1 expression levels can affect cartilage degradation by modifying Wnt signaling showing its role in maintaining joint health.
