Supplementary information

WISP2 also known as WNT1-inducible signaling pathway protein 2 or CCN5 is a protein that weighs approximately 29 kDa. This protein belongs to the CCN family which includes connective tissue growth factor and nephroblastoma overexpressed gene. WISP2 expression appears in various human tissues particularly in the liver lung and breast tissues. It is known to engage in cellular processes often impacting cell growth and differentiation due to its role in signaling pathways.
Biological function summary

Researchers observe WISP2’s significant role in cell proliferation migration and anti-apoptotic activities. It is known to not be part of a larger protein complex. This protein influences cell behavior and works in modulating functions related to the extracellular matrix. This modulation aids cellular restructuring and organization which are essential processes in tissue repair and development.

Pathways

Studies demonstrate that WISP2 plays a role in the Wnt signaling and PI3K/Akt pathways. The presence of WISP2 in these pathways allows it to control signals that regulate cell growth and survival. Within these pathways WISP2 interacts with several proteins including beta-catenin and AKT1 which help in transmitting signals from cell surface receptors to the DNA in the nucleus influencing gene expression and cellular responses.

WISP2 shows relevance in cancer and fibrosis. Researchers study its expression in breast cancer noting that it impacts tumor development and progression by interacting with other proteins like estrogen receptor-alpha (ERα). Moreover in the context of fibrosis WISP2 may influence fibrotic processes possibly involving connective tissue growth factor (CTGF) which contributes to tissue scarring and thickening through extracellular matrix production.
