Immunohistochemical analysis of rat bladder tissue, staining Wnt10a with ab106522.

Tissue was fixed with formalin and blocked with 5000 μg/ml BSA for 30 minutes at 22°C; antigen retrieval was by heat mediation in citrate buffer (pH 6). Samples were incubated with primary antibody (1/100 in BSA) for 1 hour at 22°C. An AlexaFluor®555-conjugated goat anti-rabbit polyclonal IgG (1/400) was used as the secondary antibody.

Wnt10a also known as Wingless-type MMTV integration site family member 10A is a protein that plays an important role in cellular signaling. This protein has a molecular mass of approximately 46 kDa. Wnt10a is expressed in various tissues including skin teeth and hair follicles where it is involved in mediating several cellular processes. The protein participates in signaling events by binding to specific receptors on the cell surface initiating a cascade of intracellular activities that influence development and cell differentiation.
Biological function summary

The Wnt10a protein is involved in the regulation of development and differentiation of ectodermal organs. It acts as a ligand in the Wnt signaling pathway which is a critical component of developmental processes. Wnt10a does not form complexes with others but binds to Frizzled receptors on the cell surface to propagate signals inside the cell. These signaling events guide proper formation and functioning of skin hair teeth and certain other structures influencing tissue maintenance and growth.

Wnt10a plays a major role in the Wnt signaling pathways specifically the canonical Wnt/β-catenin pathway.
