Supplementary information

Wnt10b also known as wingless-related integration site 10b is a protein involved in signaling pathways critical for development and cellular processes. It has a molecular mass of approximately 46 kDa. This protein is mainly expressed in tissues like skin lung and adipose tissue. Wnt10b belongs to the broader Wnt family which is involved in mediating several signaling events during development.
Biological function summary

Wnt10b influences cell fate decisions and plays a role in adipogenesis and osteogenesis. It is not standalone but part of the Wnt signaling complex involving interactions with other proteins such as Frizzled receptors and co-receptors like LRP5/6. Through these interactions Wnt10b influences the beta-catenin pathway impacting gene transcription and cellular differentiation.

Pathways

Wnt10b plays roles in the Wnt/beta-catenin and Wnt/JNK pathways. These pathways control embryonic development and are important for tissue homeostasis. Wnt10b activates beta-catenin which partners with TCF/LEF transcription factors to modulate the expression of target genes. This protein shows functional similarity and connectivity with other Wnt family members such as Wnt3a which also engages in beta-catenin signaling.

Wnt10b has associations with obesity and osteoporosis. Elevated Wnt10b expression can inhibit adipogenesis linking it to potential obesity resistance. Conversely dysregulated Wnt signaling including Wnt10b may impair bone density and result in osteoporosis. The protein's interaction with LRP5 is significant in bone mass regulation highlighting Wnt10b's role in these conditions.

Target data

Member of the Wnt ligand gene family that encodes for secreted proteins, which activate the Wnt signaling cascade. Specifically activates canonical Wnt/beta-catenin signaling and thus triggers beta-catenin/LEF/TCF-mediated transcriptional programs. Involved in signaling networks controlling stemness, pluripotency and cell fate decisions. Acts in the immune system, mammary gland, adipose tissue, bone and skin.
