Wnt16 also known as wingless-type MMTV integration site family member 16 is a protein with a mass of approximately 43 kDa. It belongs to the Wnt protein family which comprises secreted signaling proteins. Wnt16 facilitates the transfer of signals between cells influencing various cellular processes. This protein shows expression in several tissues such as bone cartilage and hematopoietic tissues playing roles in maintaining their functions.
Biological function summary

Wnt16 influences bone development and maintenance by enhancing osteoblast proliferation and differentiation. It functions as part of a complex signaling cascade interacting with receptors on cell surfaces to initiate intracellular signaling pathways. One of its primary roles is in balancing bone formation and resorption ensuring skeletal integrity. Furthermore Wnt16 contributes to cartilage homeostasis preventing degenerative changes that could lead to diseases like osteoarthritis.

Pathways

Wnt16 is part of significant signaling pathways like the canonical Wnt/β-catenin pathway as well as the non-canonical Wnt pathways. In the canonical pathway Wnt16 binds to Frizzled receptors and LRP5/6 co-receptors facilitating the stabilization and translocation of β-catenin into the nucleus affecting gene transcription. In this context Wnt16 relates closely to other Wnt proteins like Wnt3a influencing diverse cellular outcomes such as proliferation and differentiation.

Wnt16 has an impact on conditions like osteoporosis and osteoarthritis. In osteoporosis the reduction in Wnt16 expression correlates with decreased bone density and increased risk of fractures. Through its interaction with bone-forming proteins like Osteocalcin Wnt16 regulates bone mass and strength. Additionally in osteoarthritis Wnt16 downregulation leads to increased cartilage degradation linking it to proteins like MMP-13 which is involved in cartilage breakdown.
