Immunoprecipitation - Anti-Wnt16 antibody [EPR22889-129] (AB269278)
Wnt16 was immunoprecipitated from 0.35 mg HEK-293 (human embryonic kidney epithelial cell) whole cell lysate using ab269278 at 1/30 dilution. Western blot was performed on the immunoprecipitate using ab269278 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP)(ab131366) was used as the secondary anyibody at 1/5000 dilution.

The expression profile observed is consistent with what has been described in the literature, the band of 45 KDa represents the full length Wnt16, the band of 32 KDa represents the degraded form of Wnt16. (PMID : 26254351, 17200136).
Negative control : U-2 OS (PMID : 25999350).
This blot was developed using a higher sensitivity ECL substrate.

It functions as part of a complex signaling cascade interacting with receptors on cell surfaces to initiate intracellular signaling pathways. One of its primary roles is in balancing bone formation and resorption ensuring skeletal integrity. Furthermore Wnt16 contributes to cartilage homeostasis preventing degenerative changes that could lead to diseases like osteoarthritis.
Pathways
Wnt16 is part of significant signaling pathways like the canonical Wnt/β-catenin pathway as well as the non-canonical Wnt pathways. In the canonical pathway Wnt16 binds to Frizzled receptors and LRP5/6 co-receptors facilitating the stabilization and translocation of β-catenin into the nucleus affecting gene transcription. In this context Wnt16 relates closely to other Wnt proteins like Wnt3a influencing diverse cellular outcomes such as proliferation and differentiation.
