Wnt2 also known as Wingless-type MMTV integration site family member 2 or IRP is a protein around 41 kDa in mass. It is mechanically involved in signaling processes playing an important role in cell-to-cell communication. Wnt2 is expressed in various tissues including the heart lung and spleen and it participates in embryonic development and organogenesis. This protein belongs to the Wnt family which is known for modulating developmental processes and signaling pathways.
Biological function summary

With respect to the Wnt/β-catenin signaling pathway Wnt2 operates as a ligand that binds to Frizzled receptors on the cell surface. This interaction leads to the stabilization of β-catenin in the cytoplasm impacting gene transcription. Wnt2 does not function alone; it forms complexes with other proteins to efficiently propagate signals that regulate cell fate decisions. It is important in maintaining homeostasis and regeneration processes in adults.

Pathways

Within the Wnt/β-catenin pathway Wnt2 activation regulates important genes involved in differentiation and proliferation. This protein interacts with key molecules like Axin and GSK-3β facilitating the stabilization of β-catenin and subsequent gene transcription. Another pathway involving Wnt2 is the planar cell polarity pathway where it influences cytoskeletal arrangements and directional cell movement through its interaction with Dishevelled proteins.

Research links Wnt2 to cancer progression and fibrotic diseases. Aberrant Wnt2 signaling promotes tumorigenesis through deregulated cell proliferation and survival particularly in gastric and colorectal cancers. In fibrosis overactive Wnt2 signaling contributes to excessive extracellular matrix production with Transforming Growth Factor-β (TGF-β) often collaborating in these processes to exacerbate disease progression. Understanding how Wnt2 interacts with these proteins offers insight into potential therapeutic targets.
