Supplementary information

Wnt4 also known as Wingless-type MMTV integration site family member 4 is a protein that has significant role in cell signaling. With a molecular mass of approximately 39 kDa this protein is expressed in various tissues notably in mammalian systems such as human thyroid tissue. Wnt4 primarily facilitates communication between cells by activating signaling pathways that regulate development and differentiation processes. It is widely studied in different species including model organisms like the marmoset.
Biological function summary

Wnt4 is integral to embryonic development influencing the formation of organs and tissues. As part of the Wnt signaling pathway it works as a ligand interacting with Frizzled receptors on the cell surface. Wnt4 often in collaboration with other Wnt proteins contributes to cellular processes like proliferation migration and gene expression modulation. Its function during the development of reproductive systems and kidney morphogenesis highlights its importance in developmental biology.

Pathways

Wnt4 operates within the canonical and non-canonical Wnt signaling pathways. It primarily influences the beta-catenin signaling pathway essential for maintaining cellular homeostasis and regulating developmental signals. It can interact with other Wnt pathway components such as beta-catenin and GSK3 to modulate transcriptional activities important for cellular differentiation. By doing so it plays a pivotal role in coordinating complex signaling networks that oversee cell fate decisions.

Wnt4 displays notable associations with conditions such as Mayer-Rokitansky-Küster-Hauser (MRKH) syndrome and polycystic ovary syndrome (PCOS). Both disorders relate to abnormalities in the reproductive system highlighting Wnt4's role in sexual development. Altered Wnt4 expression can affect other signaling proteins like beta-catenin contributing to disease pathology. By understanding its contribution to these disorders researchers aim to uncover potential therapeutic targets that rectify imbalances in Wnt signaling.

Target data

Ligand for members of the frizzled family of seven transmembrane receptors (Probable). Plays an important role in the embryonic development of the urogenital tract and the lung (PubMed : 15317892, PubMed : 16959810, PubMed : 18179883, PubMed : 18182450). Required for normal mesenchyme to epithelium transition during embryonic kidney development. Required for the formation of early epithelial renal vesicles during kidney development (By similarity). Required for normal formation of the Mullerian duct in females, and normal levels of oocytes in the ovaries (PubMed : 15317892, PubMed : 16959810, PubMed : 18182450). Required for normal down-regulation of 3 beta-hydroxysteroid dehydrogenase in the ovary (PubMed : 15317892, PubMed : 16959810, PubMed : 18182450). Required for normal lung development and for normal patterning of trachael cartilage rings (By similarity).
