Wnt8b known as Wingless-type MMTV integration site family member 8B is a secreted protein involved in the Wnt signaling pathway. Its molecular weight is approximately 37 kDa. It is expressed in several tissues including the developing central nervous system and somites during embryogenesis. Wnt8b belongs to the family of Wnt proteins which play a role in cell communication and gene expression regulation.
Biological function summary

Wnt8b has a significant role in the regulation of cell fate and patterning during development. It acts as a morphogen influencing the gradient-mediated processes that define tissue differentiation. As part of the canonical Wnt signaling pathway Wnt8b binds to the Frizzled receptors and co-receptors like LRP5/6 leading to stabilization of the beta-catenin complex in the cytoplasm. This beta-catenin complex translocates to the nucleus where it affects the transcription of target genes.

Pathways

Wnt8b is involved in two major signaling pathways: the canonical Wnt/beta-catenin pathway and the planar cell polarity pathway. In the Wnt/beta-catenin pathway Wnt8b interacts with proteins like Dishevelled affecting downstream effectors such as beta-catenin. In the planar cell polarity pathway Wnt8b influences cellular orientation and movement through interactions with proteins such as Rho and Rac contributing to tissue polarity and motility.

Wnt8b is associated with conditions like cancer and neurodevelopmental disorders. Aberrant Wnt8b expression can lead to tumor progression and metastasis with connections to oncogenes such as MYC and CTNNB1. In neurodevelopmental disorders disruptions in Wnt8b signaling can impact brain development and function relating to proteins that mediate synaptic communication and plasticity.
