Non-transfected (–) and transfected (+) 293T whole cell extracts (30 μg) were separated by 10% SDS-PAGE, and the membrane was blotted with Wnt8b antibody [HL1549] (ab308168) diluted at 1 : 5000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.

Biological function summary
Wnt8b has a significant role in the regulation of cell fate and patterning during development. It acts as a morphogen influencing the gradient-mediated processes that define tissue differentiation. As part of the canonical Wnt signaling pathway Wnt8b binds to the Frizzled receptors and co-receptors like LRP5/6 leading to stabilization of the beta-catenin complex in the cytoplasm. This beta-catenin complex translocates to the nucleus where it affects the transcription of target genes.
Pathways
Wnt8b is involved in two major signaling pathways: the canonical Wnt/beta-catenin pathway and the planar cell polarity pathway. In the Wnt/beta-catenin pathway Wnt8b interacts with proteins like Dishevelled affecting downstream effectors such as beta-catenin. In the planar cell polarity pathway Wnt8b influences cellular orientation and movement through interactions with proteins such as Rho and Rac contributing to tissue polarity and motility.
