Supplementary information

XPNPEP3 also known as X-prolyl aminopeptidase 3 is a mitochondrial enzyme with a mass of approximately 55 kDa. This enzyme specifically cleaves N-terminal proline from peptides playing an important role in peptide metabolism. XPNPEP3 is widely expressed with notable presence in kidney and skeletal muscle tissues. Researchers frequently study its role in cellular processes due to its involvement in the regulation of peptide-related functions.
Biological function summary

XPNPEP3 participates in maintaining cellular homeostasis by processing peptides for further cellular activities. This enzyme works as an independent entity and is not typically part of a larger protein complex. Its role in proteolysis helps mediate cellular responses to metabolic stress by breaking down specific target peptides ensuring proper peptide recycling and utilization within cells. These functions contribute to overall organismal health and metabolic efficiency.

Pathways

The enzyme XPNPEP3 takes part in the mitochondrial processing pathway which involves the degradation and modification of mitochondrial peptides. It is closely linked to the activity of other mitochondrial enzymes such as MPP (mitochondrial processing peptidase). XPNPEP3's actions fit within the broader context of the mitochondrial function impacting pathways responsible for energy production and mitochondrial homeostasis ensuring proper cellular functionality.

Studies associate XPNPEP3 with nephronophthisis-like kidney disorder and related ciliopathies. Mutations affecting XPNPEP3 expression or function can lead to improper peptide processing contributing to the pathogenesis of these conditions. The enzyme operates alongside proteins involved in renal development and function such as NEK8 highlighting its important role in maintaining healthy kidney tissue. Understanding XPNPEP3's contributions can offer insights into potential therapeutic approaches for such disorders.

Target data

Catalyzes the removal of a penultimate prolyl residue from the N-termini of peptides, such as Leu-Pro-Ala (PubMed : 25609706, PubMed : 28476889). Also shows low activity towards peptides with Ala or Ser at the P1 position (PubMed : 28476889).. Isoform 1. Promotes TNFRSF1B-mediated phosphorylation of MAPK8/JNK1 and MAPK9/JNK2, suggesting a function as an adapter protein for TNFRSF1B; the effect is independent of XPNPEP3 peptidase activity. May inhibit apoptotic cell death induced via TNF-TNFRSF1B signaling.
