YAF2 also known as YY1AP1 is a protein that functions mechanically by interacting with other proteins to influence gene transcription. It has a molecular mass of approximately 19 kDa. YAF2 is expressed in a wide range of tissues; notable expression can be found in the nucleus where it can modulate the function of other nuclear factors. It associates particularly with regions of chromatin and is involved in the regulation of transcriptional activity.
Biological function summary

YAF2 plays a role in regulating cell proliferation and apoptosis. It is part of the polycomb group complex which modifies chromatin structure and directly impacts gene expressivity. YAF2 interfaces with proteins like YY1 functioning as a transcription cofactor. It modulates gene expression by either repressing or enhancing transcription depending on the context and specific gene targets being acted upon.

Pathways

This protein is involved in the Wnt signaling pathway and the cell cycle regulation pathway. In the Wnt signaling pathway YAF2 influences the transcriptional regulation of target genes by impacting cellular growth and differentiation processes. It is also related to proteins within these pathways such as β-catenin in the Wnt pathway where it can potentially modulate downstream effects. Through regulating the cell cycle YAF2 impacts the transition between different cell cycle stages interfacing with proteins like cyclins that control progression through the cell cycle.

Alterations in YAF2 activity relate to cancer and developmental disorders. Aberrant expression or mutations in YAF2 have been linked to oncogenic processes particularly in the context of its interactions with YY1 which is a known factor involved in various cancers. Additionally disruptions in its role within the polycomb group complex may contribute to developmental impairments affecting how gene silencing is orchestrated during embryogenesis. Further investigations into YAF2 and its interactions could provide insights into therapeutic targets for these conditions.

Component of the E2F6.com-1 complex, a repressive complex that methylates 'Lys-9' of histone H3, suggesting that it is involved in chromatin-remodeling.
