ZBTB1 also known as Zinc Finger and BTB Domain-Containing Protein 1 is a transcriptional regulator with a mass of approximately 91 kDa. It contains a BTB (Broad-Complex Tramtrack Bric-à-brac) domain and several zinc finger motifs which are common features for DNA binding and protein-protein interactions. ZBTB1 is mainly expressed in hematopoietic tissues including the thymus and spleen where it influences the development and function of immune cells.
Biological function summary

ZBTB1 plays a vital role in the regulation of gene expression during immune cell differentiation and development. It is part of a protein complex that helps to repress or activate specific target genes influencing the maturation of T cells and B cells. In addition ZBTB1 regulates the expression of genes involved in cell cycle control and apoptosis impacting immune homeostasis and cellular responses to stress.

Pathways

Researchers place ZBTB1 in significant pathways involved with immune system regulation and hematopoiesis. It interacts with proteins in the Notch signaling pathway a critical pathway for T cell development. ZBTB1 also connects with the Wnt signaling pathway where it interacts with other zinc finger proteins to modulate gene expression during early immune cell differentiation.

ZBTB1 mutations have been associated with immunodeficiencies and certain types of cancer such as T-cell acute lymphoblastic leukemia (T-ALL). The protein's connection to improper immune cell differentiation links it to autoimmune disorders as well. Additionally ZBTB1 interacts with tumor suppressor proteins like TP53 influencing cell cycle regulation and apoptosis factors that are often dysregulated in cancers.
