Supplementary information

The ZBTB38 protein also known as CIBZ (CTCFL interacting BTB zinc finger protein) functions as a transcriptional regulator with a molecular mass of approximately 149 kDa. It contains zinc finger motifs that bind to DNA sequences to modulate gene expression. ZBTB38 mostly localizes in the nucleus of cells and shows expression in human tissues such as brain heart and skeletal muscle. The presence in these tissues indicates an important role in fundamental cellular processes.
Biological function summary

The regulatory role of ZBTB38 impacts cell cycle control and differentiation. It participates in transcriptional repression or activation depending on the interacting partners. Known to form part of multisubunit complexes ZBTB38 can engage in molecular interactions that influence chromatin remodeling. Through these configurations it affects the transcriptional regulation of target genes influencing cellular functions and development pathways.

Pathways

ZBTB38 is involved in the regulation of the Wnt and MAPK signaling pathways. These pathways play a significant role in cell proliferation differentiation and survival. ZBTB38 directly interacts with proteins such as CTCF (CCCTC-binding factor) and CTCFL (CTCF-like) in these pathways influencing various cellular responses. The precise modulation of these pathways by ZBTB38 contributes to maintaining cellular homeostasis and dynamics.

Disruptions in ZBTB38 function are linked to certain cancers and developmental disorders. Abnormal expression or mutations in ZBTB38 are associated with an increased risk of neuroblastoma where it interacts with MYC a known oncogene. Additionally changes in ZBTB38 are connected with developmental syndromes possibly through its interactions with CTCF impacting gene expression profiles relevant to these conditions.

Target data

Transcriptional regulator with bimodal DNA-binding specificity. Binds with a higher affinity to methylated CpG dinucleotides in the consensus sequence 5'-CGCG-3' but can also bind to E-box elements (5'-CACGTG-3'). Can also bind specifically to a single methyl-CpG pair. Represses transcription in a methyl-CpG-dependent manner (PubMed : 16354688). Plays an important role in regulating DNA replication and common fragile sites (CFS) stability in a RBBP6- and MCM10-dependent manner; represses expression of MCM10 which plays an important role in DNA-replication (PubMed : 24726359). Acts as a transcriptional activator. May be involved in the differentiation and/or survival of late postmitotic neurons (By similarity).
