ZBTB4 also known as Zinc Finger and BTB Domain-Containing Protein 4 is a transcriptional repressor with a mass of approximately 140 kDa. ZBTB4 contains a BTB/POZ domain and several zinc finger motifs allowing it to bind DNA and suppress gene expression. It is expressed in various tissues with higher levels observed in the brain lung and liver. The protein primarily localizes in the nucleus where it regulates the transcription of specific target genes.
Biological function summary

ZBTB4 interacts with other proteins as a part of transcriptional repression complexes. Its role includes modulating gene expression linked to cellular proliferation and differentiation. ZBTB4 often works together with corepressors such as CtBP which enhances its repressive function. Through these interactions it helps maintain genomic stability and manage cell cycle progression.

Pathways

Research indicates that ZBTB4 is an important component of the p53 signaling and apoptosis pathways. Transcriptional repressor with bimodal DNA-binding specificity. Represses transcription in a methyl-CpG-dependent manner. Binds with a higher affinity to methylated CpG dinucleotides in the consensus sequence 5'-CGCG-3' but can also bind to the non-methylated consensus sequence 5'-CTGCNA-3' also known as the consensus kaiso binding site (KBS). Can also bind specifically to a single methyl-CpG pair and can bind hemimethylated DNA but with a lower affinity compared to methylated DNA (PubMed : 16354688). Plays a role in postnatal myogenesis, may be involved in the regulation of satellite cells self-renewal (By similarity).
