Supplementary information

ZCCHC8 or Zinc Finger CCHC-Type Containing 8 is a protein that stabilizes RNA and is involved in RNA decay playing a role in RNA metabolism. It is composed of approximately 92 kDa and is found mainly in the nucleus. ZCCHC8 is expressed in many tissues with notable levels in the brain and heart indicating its relevance in widely important cellular processes. Its function is not limited to RNA stabilisation as it also contributes to RNA processing and transport.
Biological function summary

ZCCHC8 participates in regulating gene expression by being a part of the nuclear exosome targeting (NEXT) complex which is responsible for the degradation of a variety of RNA species including long non-coding RNAs and promoter upstream transcripts. This involvement in RNA turnover indicates that ZCCHC8 helps maintain cellular RNA homeostasis contributing to the control of gene expression. The NEXT complex which includes other proteins like RBM7 and hMTR4 functions as a critical component in deciding which RNA substrates are selected for degradation reflecting the dynamic nature of RNA processing events in cells.

Pathways

ZCCHC8 is significant in the RNA surveillance pathway and RNA degradation pathway. Within these pathways ZCCHC8 contributes by ensuring defective or excess RNA molecules are adequately processed and degraded therefore preserving RNA quality and balance. It interacts with proteins such as RBM7 and the exosome complex emphasizing its integrative role in RNA metabolism and the importance of these interactions in safeguarding correct cellular function through efficient RNA management.

Alterations in ZCCHC8 function or expression levels have been associated with neurological disorders such as neurodevelopmental diseases and psychiatric conditions. Its connection with RNA metabolism suggests that disruptions in ZCCHC8 activity may affect neuronal function and development. Research has implicated the protein in disorders like schizophrenia where it likely interacts with RNA-binding proteins such as FMRP known to bind mRNA and is associated with fragile X syndrome. These associations highlight ZCCHC8's role in maintaining neural integrity and mental health.

Target data

Scaffolding subunit of the trimeric nuclear exosome targeting (NEXT) complex that is involved in the surveillance and turnover of aberrant transcripts and non-coding RNAs (PubMed : 27871484). NEXT functions as an RNA exosome cofactor that directs a subset of non-coding short-lived RNAs for exosomal degradation. May be involved in pre-mRNA splicing (Probable). It is required for 3'-end maturation of telomerase RNA component (TERC), TERC 3'-end targeting to the nuclear RNA exosome, and for telomerase function (PubMed : 31488579).
