Immunohistochemical analysis of formalin fixed paraffin embedded human pancreas tissue labelling ZDHHC4/DC1 with ab235369 at a 1 : 50 dilution. The immunostaining was performed on a Leica Bond™ system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum for 30 min at RT. ab235369 was incubated at 4°C overnight. The primary was detected using a Goat anti-rabbit polymer IgG HRP and visualized using 0.05% DAB. Secondary antibody only control : uses 1% BSA instead of primary antibody

Biological function summary
This palmitoyltransferase influences protein-protein interactions by adding lipid moieties to key signaling molecules. It is part of the ZDHHC family of proteins which collaborate to modify a variety of substrates important for cellular signaling pathways. By palmitoylating proteins ZDHHC4 can regulate pathways involving signal transduction and vesicular trafficking affecting how signals are transmitted inside the cell and how proteins are transported to different cellular compartments.
Pathways
Research shows this enzyme acts within the signaling pathways such as the Wnt signaling and Ras signaling. In the Wnt pathway ZDHHC4's palmitoylation activity helps control the stability and localization of proteins important for signal propagation. Within the Ras signaling pathway it modifies Ras proteins which are related to cell growth and survival. These associations highlight its role in influencing cell behavior through complex signaling networks.
