ZEB2 also known as SIP1 (Smad-interacting protein 1) is a zinc finger E-box-binding homeobox 2 protein. It has a mass of approximately 150 kDa. ZEB2 functions as a transcriptional repressor in cellular processes. This protein can bind to E-box sequences in DNA regulating gene expression by repressing transcription. ZEB2 expresses mainly in neural crest-derived tissues the central nervous system and the developing heart and plays an important role in these areas.
Biological function summary

ZEB2 influences epithelial-mesenchymal transition (EMT) and acts as a regulator in developmental processes. It is part of a protein complex with Smad proteins which are key transducers of the transforming growth factor-beta (TGF-β) signaling pathway. ZEB2 modulates the expression of genes involved in differentiation proliferation and cell migration which are essential for tissue development and response to environmental cues.

Pathways

ZEB2 plays roles in the TGF-β and Wnt signaling pathways. It interacts with other proteins such as Smad2/3 in the TGF-β pathway to control gene expression related to EMT and cell differentiation. In the Wnt signaling pathway ZEB2 influences the regulation of genes that mediate cell proliferation and adhesion. These pathways are vital for normal development and homeostasis making ZEB2 an important player in cellular function.

ZEB2 has links to Mowat-Wilson syndrome and cancer progression. In Mowat-Wilson syndrome mutations in the ZEB2 gene lead to neurodevelopmental abnormalities. ZEB2 also contributes to cancer by facilitating EMT which is involved in tumor invasion and metastasis. It connects with other EMT-related proteins such as Snail and Slug in cancer contexts emphasizing its importance in disease mechanisms.
