Samples : Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from MDA-MB-435 cells prepared using NETN lysis buffer. Antibodies : Rabbit antiZEB2 recombinant monoclonal antibody [BLR115H] used for IP at 20 µl/mg lysate. ZEB2 was also immunoprecipitated by rabbit anti-ZEB2 antibodies. For blotting immunoprecipitated ZEB2, A700-115 was used at 1 : 1000. Chemiluminescence with an exposure time of 3 minutes.

Biological function summary
ZEB2 influences epithelial-mesenchymal transition (EMT) and acts as a regulator in developmental processes. It is part of a protein complex with Smad proteins which are key transducers of the transforming growth factor-beta (TGF-β) signaling pathway. ZEB2 modulates the expression of genes involved in differentiation proliferation and cell migration which are essential for tissue development and response to environmental cues.
Pathways
ZEB2 plays roles in the TGF-β and Wnt signaling pathways. It interacts with other proteins such as Smad2/3 in the TGF-β pathway to control gene expression related to EMT and cell differentiation. In the Wnt signaling pathway ZEB2 influences the regulation of genes that mediate cell proliferation and adhesion. These pathways are vital for normal development and homeostasis making ZEB2 an important player in cellular function.
