ZFHX4 also known as zinc finger homeobox 4 is a transcription factor involved in regulating gene expression. It has a molecular mass of approximately 384 kDa. This protein is large and contains multiple zinc finger domains allowing it to bind DNA and influence transcription of specific genes. ZFHX4 is widely expressed in human tissues with higher expression noted in the brain and eyes suggesting its important role in neural function and ocular development.
Biological function summary

ZFHX4 plays a role in neural development and maintenance. This transcription factor is not known to be part of a larger protein complex but it interacts with several other proteins to control neuronal differentiation and maturation. In the retina ZFHX4 influences the growth and specialization of neural cells. This role in cellular development highlights its importance in maintaining the normal function of neural tissues.

Pathways

ZFHX4 influences both the Wnt signaling and Notch signaling pathways which are critical in development and cellular differentiation. Within these pathways it interacts with proteins such as β-catenin in the Wnt pathway and Notch receptors in the Notch pathway. These interactions ensure proper functioning of the pathways facilitating cell proliferation and differentiation during development.

ZFHX4 associates with ocular diseases such as Axenfeld-Rieger syndrome and corneal dystrophy. Mutations or dysregulation of ZFHX4 can lead to developmental defects in the eye. In Axenfeld-Rieger syndrome ZFHX4 interacts with other transcription factors like FOXC1 affecting the development of the anterior segment of the eye. In corneal dystrophy the interruption in its regulation can lead to abnormal corneal cell function and vision impairment.
