This data was developed using ab314323, the same antibody clone in a different buffer formulation. ZFYVE27 was immunoprecipitated from 0.35 mg SH-SY5Y (human neuroblastoma epithelial cell) whole cell lysate with ab314323 at 1/30 dilution (2ug in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab314323 at 1/1000 dilution. VeriBlot for IP secondary antibody(HRP)(ab131366) was used at 1/5000 dilution. Blocking and diluting buffer and concentration : 5% NFDM/TBST Lysates in lane 1-3 were freshly made and used immediately to minimize protein degradation. ZFYVE27 also known as Protrudin functions as an active component in intracellular transport processes. The protein has an approximate molecular weight of 50 kDa. It is widely expressed in various tissues with significant levels found in the brain where it interacts with other proteins to facilitate its role in cellular dynamics. ZFYVE27 contains a FYVE domain which aids in binding to phosphatidylinositol 3-phosphate at endosomal membranes supporting its function in membrane trafficking.
Biological function summary

ZFYVE27 plays a role in neurite extension and cellular trafficking. It is not part of a larger complex but interacts independently with other proteins to regulate vesicle formation and transport along microtubules. ZFYVE27 helps in the maintenance of the endoplasmic reticulum's structure and function. Additionally it interacts with proteins like kinesin which assist in the movement of cargo along cellular structures important for proper cell function and communication.

Pathways

ZFYVE27 contributes significantly to the processes of membrane trafficking and neuronal connectivity. It is associated with the endosomal sorting pathway influencing the movement and recycling of vesicles within cells. In this pathway ZFYVE27 interacts with other proteins such as Rab11 which is essential for guiding vesicles to their intended cellular destinations. Additionally it has a role in the axon growth pathway where it cooperates with kinesin motor proteins to promote the elongation and maintenance of neuronal projections.

ZFYVE27 has a connection with hereditary spastic paraplegia (HSP). Mutations in the ZFYVE27 gene lead to neural structural abnormalities contributing to the formation of this neurological disorder characterized by progressive limb spasticity and weakness. The protein interacts with spastin another protein associated with HSP with implications for axonal regeneration and stability. ZFYVE27 also holds a potential link with certain neurodegenerative conditions given its important role in maintaining neuronal network integrity.

Promotes axonal elongation and contributes to the establishment of neuronal cell polarity (By similarity). Involved in nerve growth factor-induced neurite formation in VAPA-dependent manner (PubMed : 19289470). Contributes to both the formation and stabilization of the tubular ER network (PubMed : 24668814). Involved in ER morphogenesis by regulating the sheet-to-tubule balance and possibly the density of tubule interconnections (PubMed : 23969831).
