Zinc Fingers and Homeoboxes 1 (ZHX1) known also as ZNF908 is a zinc finger-containing protein with a molecular mass of roughly 97 kDa. Consisting of three C2H2-type zinc fingers and five homeodomains it functions primarily as a transcriptional repressor. ZHX1 is expressed in a wide range of tissues including the liver kidney and spleen. Through its ability to dimerize ZHX1 interacts with DNA regulating gene transcription by binding to specific promoters.
The protein plays a role in modulating gene expression by forming homodimers or heterodimers with other transcription factors such as ZHX2 and ZHX3. ZHX1 represses the transcription of target genes that are critical for cell growth and differentiation. In this process ZHX1 can join in a multi-protein complex further influencing transcriptional activity. This multifaceted role signifies its involvement in maintaining normal cellular function and growth.
ZHX1 exerts influence within the calcium signaling pathway and cell cycle regulation. It participates in pathways that control cellular proliferation by interacting with proteins such as cyclins and cyclin-dependent kinases. In calcium signaling ZHX1 modulates gene expression in response to calcium ion fluctuations affecting cellular processes like apoptosis and metabolism. These interactions demonstrate ZHX1's important role in regulating complex cellular responses.
Studies link ZHX1 to liver cancer and nephritic syndrome. Its interaction with proteins like Cyclin D1 contributes to the abnormal cell proliferation observed in hepatocellular carcinoma. In nephritic syndrome altered expression of ZHX1 affects albumin synthesis influencing disease progression. Understanding these connections highlights ZHX1 as a potential biomarker and therapeutic target in these conditions.
