Zika virus NS4B sometimes called NS4B nonstructural protein is an integral membrane protein with a mass of approximately 27 kDa. The protein localizes to the endoplasmic reticulum (ER) of infected cells. NS4B functions as an important component in the replication of the viral RNA genome. During viral replication NS4B alters the host cell membrane creating structures that favor viral replication. This protein also plays a role as a suppressor of the host's antiviral immune responses by interfering with the signaling pathways that control cytokines and interferons.
Biological function summary

NS4B interacts closely with other nonstructural proteins to form the viral replication complex a multi-protein assembly essential for efficient RNA synthesis. NS4B is known to modify the lipid composition of the ER membrane facilitating the formation of replication platforms essential for viral RNA synthesis. The protein also modulates cellular stress responses helping the virus to evade host immune surveillance. NS4B factors heavily in viral strategy to manipulate host cell processes ensuring successful viral replication and infection propagation.

Pathways

NS4B influences the interferon signaling pathway and the unfolded protein response. It suppresses interferon production impacting the interferon-stimulated gene (ISG) response by modulating the host's immune signaling cascades. NS4B directly interacts with proteins such as NS4A and NS5 contributing towards the viral RNA polymerase activity. Its interactions stabilize the replication complex enhancing RNA genome synthesis. By inhibiting the unfolded protein response NS4B affects protein homeostasis allowing the virus to continue exploiting host resources.

NS4B's function relates to Zika virus infection and it has associations with microcephaly in newborns. The protein plays a role in the virus’s pathogenic effects especially in neural cells contributing to neurodevelopmental disorders. Additionally NS4B impacts Guillain-Barré syndrome development linking it with immune-mediated damage to peripheral nerves. The interaction of NS4B with other proteins like NS1 helps modulate immune evasion strategies enhancing disease progression and infection severity.
