Supplementary information

ZIP-3 also known as SLC39A3 is a protein that functions as a zinc transporter facilitating the uptake of zinc into cells. This protein belongs to the solute carrier (SLC) family and typically has a mass of approximately 47 kDa. ZIP-3 localizes mainly to the plasma membrane and is expressed in a variety of tissues including liver kidney and intestine. It plays an important role in maintaining intracellular zinc homeostasis critical for various cellular functions.
Biological function summary

Zinc is essential for numerous cellular processes and ZIP-3 helps regulate its availability. Zinc acts as a cofactor for over 300 enzymes and plays a vital role in transcription factor function gene expression and cell signaling. Through these actions ZIP-3 impacts cellular proliferation differentiation and survival. ZIP-3 does not form part of a larger complex but works closely with other zinc transporters such as those from the ZIP and ZnT families to balance zinc levels in the body.

Pathways

Researchers have associated ZIP-3 with critical processes such as the metal ion transport and cellular response to stress. By modulating zinc levels ZIP-3 interacts with pathways like the MAPK signaling pathway which is pivotal for cell growth and division. In these pathways ZIP-3 operates alongside proteins like ZIP-4 and ZnT-1 ensuring proper zinc redistribution and cellular adaptation to changing environmental conditions.

Scientists have linked ZIP-3 dysfunction to conditions such as type 2 diabetes and neurodegenerative disorders. Abnormal ZIP-3 activity can disrupt zinc homeostasis contributing to insulin resistance in diabetes. In neurodegenerative diseases impaired zinc transport by ZIP-3 can lead to neuronal damage and cognitive deficits. Research also examines the interplay of ZIP-3 with other proteins such as insulin receptor substrate (IRS) in diabetes highlighting the potential for targeted therapeutic interventions.
